The composition depicts Saint Jerome, a central figure in Christian iconography, situated within a rugged, rocky wilderness. The saint is shown in a state of quiet contemplation, his posture suggesting a moment of intellectual or spiritual focus. He is draped in a pink cloth, which provides a distinct colour contrast against the muted, earthy tones of the surrounding stone formations and the shadowed forest.

Lotto employs a dramatic use of light, which illuminates the saint and the immediate rock face while leaving the periphery in deeper shadow. This technique draws the viewer's eye directly to the figure of Jerome and his open book. The background reveals a glimpse of a distant, atmospheric horizon, typical of the Venetian school's interest in natural light and atmospheric perspective. The craggy, vertical rock formations dominate the upper portion of the panel, creating a sense of isolation and enclosure that reflects the ascetic life of the saint.

Unlike many contemporary depictions of the same subject, which often include a lion or cardinal's hat, this version focuses on the solitary act of meditation.
